Antibody purification
Polyclonal antibodies are purified from hyperimmune serum by salt precipitation followed by ion exchange chromatography (sheep) or affinity chromatography on Protein A-Sepharose (rabbit). Specific Antibodies directed to the target antigen are purified by immunoadsorption.
Monoclonal antibodies are purified from cell culture supernatant either by precipitation or ultrafiltration followed by affinity chromatography on Protein A-Sepharose or ion exchange chromatography.
Purity is checked by PAGE, reactivity by solid-phase ELISA.
